Recent studies have identified ANK3 as a leading BD risk locus[3]. The gene has also been implicated in other neuropsychiatric disorders such as autism spectrum disorder[3]. ANK3 is involved in axon physiology, which provides a plausible mechanism for its role in BD and schizophrenia[1]. ANK3 is also involved in the maintenance of protein location in the plasma membrane, membrane assembly, mitotic cytokinesis, negative regulation of delayed rectifier potassium channel activity, and neuromuscular junction development[2]. Studies have found that ANK3 is associated with cognitive performance and cortical thickness in patients with first-episode psychosis[1]. The gene has been implicated in risk of stress-related and externalizing disorders beyond its previous associations with BD and schizophrenia[2]. ANK3 has been found to be involved in psychotic experiences across development and shared between psychotic experiences and psychiatric disorders[4].
